چکیده

Colicins Ia and Ib have been purified from mitomycin C-induced Escherichia coli strains W3110 str-r (co1 Ia) and W3110 str-r (co1 Ib) by ammonium sulfate precipitation, treatment with diethylaminoethylcellulose, and chromatography on DEAE-Sephadex, phosphocellulose, and hydroxylapatite. Sedimentation velocity and equilibrium centrifugation as well as polyacrylamide gel electrophoresis at several pH values indicated that both colicin preparations were homogeneous. The molecular weight of colicin Ia was determined from its &I,~ (3.57 S) and D&+ (4.12 X 1OF cm2 set-r) as well as from sedimentation equilibrium centrifugation and dodecyl sulfate polyacrylamide gel electrophoresis to be approximately 77,600. The molecular weight of colicin Ib was approximately 79,600 (a!&, = 3.67 S; D&W = 4.32 x lo-’ cm2 set-l). Colicins Ia and Ib had f/j0 values of 1.82 and 1.76, respectively.

Characterization of colicin Ia and colicin Ib. Chemical studies of protein structure.

The chemical properties of purified colicin Ia-CA53 and colicin Ib-P9 have been compared. Colicins Ia and Ib are basic proteins having isoelectric points of greater than 10 and 9.5, respectively. The colicins exhibit similar amino acid compositions having a high content of basic amino acids and low amounts of hydrophobic residues. Neither colicin contains cysteine. Both colicins have serine as ...

Detection of Colicin genes by PCR in Escherichia coli isolated from cattle in Shiraz-Iran

A variety of probiotic bacteria have been tested to control animal and foodborne pathogenic bacteria in livestock. The mechanism of inhibition of pathogenic bacteria for several of those probiotic microorganisms is mediated by the production of bacteriocins. Colicins are probably the group of bacteriocins that have been most thoroughly characterized. Colicins are antimicrobial proteins produced...

Solubilization and partial characterization of the colicin I receptor of Escherichia coli.

Treatment of the cell envelope of a colicin I-sensitive strain of Escherichia coli with the nonionic detergent Triton X-100 solubilizes an envelope component which forms a complex with 1251-colicin Ia and Ib. A similar component could not be shown in a strain known to be lacking active colicin I receptors. On this basis, this component is assumed to be the colicin I receptor. The colicin I rece...

Evidence that the immunity protein inactivates colicin 5 immediately prior to the formation of the transmembrane channel.

Determination and analysis of the nucleotide sequences of the activity, immunity, and lysis genes of colicin 5 assigned colicin 5 to the subclass of pore-forming colicins to which colicins 10, E1, Ia, Ib, and K belong. Mutational analysis of colicin 5 and exchange of DNA fragments between the most closely related colicins, colicins 5 and 10, and between their immunity proteins localized the reg...

Evolution of microcin V and colicin Ia plasmids in Escherichia coli.

Survey results and genotypic characterization of Escherichia coli strains demonstrate that the bacteriocins colicin Ia and microcin V coassociate in a strain more often than would be expected by chance. When these two bacteriocins co-occur, they are encoded on the same conjugative plasmid. Plasmids encoding colicin Ia and microcin V are nonrandomly distributed with respect to the genomic backgr...
